What's the Difference?! If you look on the back of most DVD's it says 'Mastered in High Definition'. What does that mean? Obivously it's not an HD-DVD but it's in HD? How does that make sense?!

It means the DVD-video for the disc was taken from a print of the film that was transferred to high definition, and this includes movies shot on actual film stock and not just films shot in HD. For instance, the upcoming, remastered Blade Runner: Director's Cut disc has been mastered from a high definition source by creating a digital copy of the film, cleaning it up, and creating an HD master. The DVD-video for the upcoming disc was then created by encoding an MPEG-2 version based on the HD master. Most DVDs are taken from such a source nowadays and have been for a while.
